Existing clinical studies have indicated that increased matrix stiffness is positively correlated with an unfavorable prognosis of HCC, and liver stiffness has been taken as a biomarker in clinics to predict HCC progress and its outcome. This study aims to explore whether increased matrix stiffness tuned HCC cell-released exosomal miRNAs trigger HCC metastasis. Exosomes were first isolated and purified from CM of MHCC97H cells grown on low (6 kPa) and high-stiffness (16 kPa) substrates. Small RNAs in exosomes were further subjected to sequencing via the Illumina NovaSeq 6000 platform. |
