Autophagy plays a role in the maturation of hematopoietic precursors through nucleolin pathways in the embryo |

Using the LC3-RFP-EGFP autophagy reporter mouse model, we present that the different statuses of autophagy existed in the hematopoietic precursors, correlating with hematopoietic potential. Autophagy related gene (Atg) 5 deficiency specifically in endothelial cell disrupted endothelial cell to hematopoietic transition (EHT) via blocking the fusion between autophagosome and lysosome. Using combined approaches, including single cell RNA-sequencing (scRNA-seq), we confirmed that Atg5 deletion interrupted the developmental temporal order of EHT to further affect the pre-HSC I maturation. The rescue experiments with in vivo analysis suggest that autophagy influenced the hemogenic potential of endothelial cells via nucleolin pathways. These findings show that autophagy plays vital roles in the formation / maturation of hematopoietic precursors derived from hemogenic endothelial cells. |
