Description Reactive astrocytes play important roles in brain injury, stroke, neurodegenerative diseases, and aging. Traditionally, they have been classified into neurotoxic (A1) and neuroprotective (A2) subtypes based on disease conditions and gene expression patterns. However, it remains unclear whether these transcriptomically defined substates represent final states or intermediate stages in the activation process of reactive astrocytes. In this study, we observed a consistent progression from a nonreactive state to a neuroprotective substate, and ultimately to a neurotoxic substate in astrocytes by integrating Data-independent acquisition (DIA) quantitative proteomics and transcriptomics data of primary mouse astrocytes treated with TNF+IL1a+C1q for different time points
