Description METTL3 KO in mESCs upregulates most TEs and 2C-like genes via m6A-dependent mechanisms, regardless of culture. Transient METTL3 degradation enables METTL3-deficient mESCs to contribute to trophectoderm lineages in chimeras, indicating expanded developmental potential. This demonstrates METTL3-mediated m6A modifications regulate mESC transcriptional and developmental plasticity, linking m6A loss to an extended potency state resembling the 2C-like program.
