Title Single cell RNA sequencing on synovium, meniscus, subchondral bone and cartilage of knee osteoarthritis
Description Knee osteoarthritis (OA) is one of the most common degenerative joint diseases and a major cause of pain and disability. Under single cell RNA sequencing generated by 10x Genomics, we identified fibroblast-like synoviocytes (FLSs) are the dominant communication hub, acting by releasing bone and cartilage-damaging factors such as TNF, IL6, CCL, CSF and MIF. These factors further aggravate cartilage damage and the local inflammatory response, which is markedly reduced following denosumab treatment. Further sequencing analysis identifies follistatin-like protein 1 (FSTL1) as the most upregulated cytokine in FLSs, inducing inflammatory reactions and chondrocyte catabolism, effects that are significantly inhibited by denosumab. Our results demonstrate that synovial inflammation is a critical event in the development of OA, and that FLSs could be a potential target for early intervention in OA.
