| Title | Title: Single-cell transcriptomic profiling of the primary lesions of metastatic versus non-metastatic prostate cancer patients |

| Description | The primary lesions of metastatic prostate cancer (mPCa) and non-metastatic prostate cancer (nmPCa) were investigated using single-cell RNA sequencing (scRNA-seq). The fresh tissues were washed, minced into small pieces, and then digested with 3 mL sCelLive Tissue Dissociation Solution (Singleron) at 37 °C for 15 min. The cell suspension was collected and filtered through a 40-micron sterile strainer. Afterwards, the GEXSCOPE red blood cell lysis buffer (RCLB, Singleron) was added to remove red blood cells. The mixture was incubated and then centrifuged at 300 g, 4 °C for 5 mins to remove supernatant and suspended with PBS. Single-cell suspensions were loaded onto microwell chip (Singleron). Barcoding Beads are collected from the microwell chip, followed by reverse transcription and PCR amplification. The amplified cDNA is then fragmented and ligated with sequencing adapters. The libraries were constructed with the GEXSCOPE Single Cell RNA Library Kits (Singleron) and sequenced on Illumina novaseq 6000. |
