| Description | Saliva samples were thawed at room temperature, and 100 μL was aliquoted and mixed 1:1 with 0.06 mg/mL L-2-chlorophenylalanine (internal standard). Pre-chilled MeOH:ACN was introduced, followed by 1 min of vortexing and 10 min of ice-bath sonication; proteins were then precipitated at -20 °C for 30 min.After centrifugation at 13,000 rpm, the supernatants were taken to dryness; residues were reconstituted in methanol:water (1:4, v/v), sonicated for 3 min, incubated 2h at -20 °C, and centrifuged again. The resulting supernatants underwent 0.22-µm filtration and were frozen at -80 °C until assay. QC pools were assembled from equal parts of all specimens.LC was performed on a Waters ACQUITY UPLC I-Class Plus using an ACQUITY HSS T3 column. Mobile phase A (aqueous) was 0.1% formic acid in water; mobile phase B (organic) was 0.1% formic acid in acetonitrile. |
