Description | Semigranular cells (SGCs) and granular cells (GCs) are two dominant groups of circulating hemocytes in crayfish Cherax quadricarinatus. In this study, the protein profiles of different hemocyte subpopulations and their progenitors were analyzed by two work-flows. Firstly, the cytoplasmic fractions and membrane fractions were prepared from SGCs and GCs. The protein fractions were analyzed by one-dimensional gel electrophoresis, and the cell type-specific proteins were identified by LC-MS/MS. In addition, the cytoplasmic fractions and membrane fractions were prepared from hematopoietic tissue cells, SGCs, and GCs. These protein fractions were analyzed by in-solution digestion followed by LC-MS/MS. The cell type-specific proteins/differentially expressed proteins were also screened out by this method. |
