Spike-in RNA were added to total RNA, and then RNA mixture was fragmented by sonication. Fragmented RNA was incubated with m7A antibody covered beads and washed for six times by low and high salt buffer. Then the immunoprecipitated RNA was eluted and subjected for total RNA-seq library construction pipeline. |
