The essential role of RNA polymerase I transcription in establishing the chromatin architecture in mouse embryos

Description:
Here, we found global three-dimensional genome reorganization from human oogenesis to early embryonic development, and different characteristics and mechanisms from those in mice. Furthermore, integrative analysis by clustered regularly interspaced short palindromic repeat/Cas13a gene editing, low-input genome-wide chromosome conformation capture, cleavage under targets and tagmentation, and RNA-sequencing showed that RNA polymerase I (Pol I) was essential for the de novo chromatin structure during mouse embryonic development. Mouse embryos were blocked at the 2-cell phase after knockout or inhibition of Pol I, and the biological processes related to chromatin silencing was enriched. More importantly, inhibition of Pol I transcription prevented establishment of the TAD structure in mouse embryos, but not in human embryos. Additionally, the TAD structure was weakened in mouse female germline stem cells lacking Pol I transcription, but was enhanced in human female germline stem cells.
