The flo22 mutant exhibited slower dry matter accumulation during grain filling. There was no difference in the seed setting rate. Scanning electron microscopy (SEM) of mature grains showed that starch granules in mutant endosperm cells were loosely packed and rounder, whereas endosperm cells in the wild type were fully filled with compact starch granules. Iodine staining semithin sections of developing endosperm at 9 d after flowering (DAF) showed that the mutant had smaller SGs with lots of gaps compared with the wild type. Thousand-grain weight was significantly reduced by about 17%.
